Description
Lipid peroxidation은 동, 식물에서 잘 알려진 세포손상 (cellular damage) 메카니즘으로 cell과 tissue에서 산화적 스트레스 (oxidative stress)의 indicator로서 사용됩니다.
Lipid peroxidation 과정에 의해 막 지질이 파괴되고 과산화 지질이 생성되는데 이 과산화 지질은 매우 불안정하여 reactive carbonyl compound와 같은 복잡한 화합물들로 분해됩니다.
이후 다중 불포화 지방산의 과산화물은 Malonaldehyde (Malondialdehyde, MDA) 와 같은 물질로 분해되는데 이는 과산화 지질의 대표적인 marker로 이용됩니다. Thiobarbituric Acid Reactive Substance (TBARS)의 측정은 잘 알려진 lipid peroxidation screen 방법으로, MDA가 thiobarbituric Acid (TBA)와 반응하여 형성하는 MDA-TBA Adduct를 측정함으로써 lipid peroxidation 정도를 알 수 있습니다.
이 분석은 25℃에서의 MDA와 발색 시약인 TBA의 반응에 기초하며, 산성 조건 하에서 MDA와 TBA의 반응에 의해 형성된 MDA-TBA adduct는 colorimetric 방법으로 흡광도 540 nm (530 ~ 540 nm)에서 측정합니다. (Detection limit: 1.0 μM.)
